ABSTRACT

The following experiments were designed to examine the effect of serum of spayed dogs on superovulation response in mice and rats. In Experiment 1, female mice at diestrus (n=30) were divided into three equal groups and superovulated with either administration of 5 IU pregnant mare serum gonadotropin (PMSG) or recombinant follicle stimulating hormone (rFSH) (reducing dose from 2.5 to 0.5 IU) and 5 IU human chorionic gonadotropin (hCG) administered 48h later. Serum of spayed dogs was administered intraperitoneally at a reduced dose from 0.1 to 0.025 mL in a 48 h period. In Experiment 2, female rats (n=30) at diestrus stage were divided into three equal groups. Superovulation was induced using either 30 IU PMSG, or a dose reduced from 5 to 1 IU rFSH and 25 IU hCG administered 48h later. Serum of spayed dogs was administered in a reduced dose from 0.6 to 0.1 mL in a 48 hour period. Female mice and rats were mated 24 h following hCG administration. On day 14 after mating, animals were euthanized and ovarian sections were fixed for histopathological evaluation and corpus luteum (CL) counting. No significant difference observed in mean (±SEM) number of CLs between the PMSG group and the mice that received serum of spayed dog (10.4±1.3 vs 9.2±1.0). Mean (±SEM) number of CLs tended to be lower in rats that received serum of spayed dog than those of rats which received either PMSG or rFSH (15.1±1.9 vs 23.6±3.1 and 23.1±2.9, P=0.06, respectively). In conclusion, serum of spayed dogs is able to induce a superovulatory response in mice and rats.

ABSTRACT This study aimed to determine the histological features of the endometrium of bitches, as well as the cell proliferation at specific moments of diestrus, 10, 20, 30, 40, 50 and 60 days post ovulation, correlating the endometrial thickness with the uterine cell proliferation and the metabolic state (weight, blood glucose and plasma cholesterol) of the animals. Therefore, the right and left uterine horns of 26 clinically healthy bitches submitted to ovariohysterectomy were histologically analyzed 10, 20, 30, 40, 50 and 60 days post ovulation. The hematoxylin-eosin and AgNOR staining techniques were performed. All parameters were evaluated by ANOVA and post-hoc Tukey test (p<0.05). The correlation between endometrial thickness and uterine cell proliferation, weight, blood glucose and plasma cholesterol of animals was observed using the Pearson method (p<0.05). In the present study, it is concluded that endometrial thickness does not differ at any of the moments analyzed in diestrus. The endometrial thickness is not influenced by hormones, weight, blood glucose or serum cholesterol of bitches in this phase of the estrous cycle. However, there is greater cell proliferation in the endometrium at day 40 compared to day 60 post ovulation under the influence of the endocrine profile.

ABSTRACT

A citologia uterina consiste em método prático e eficiente para o diagnóstico de afecções uterinas na égua, contribuindo para a conduta terapêutica e o prognóstico quanto à fertilidade das fêmeas. Assim, o objetivo deste trabalho foi comparar diferentes técnicas de coleta de material para citologia uterina em éguas, a fim de se verificarem possíveis diferenças qualitativas e quantitativas e a praticidade na execução das metodologias. Para tanto, 12 éguas multíparas foram submetidas ao exame citológico uterino, durante o estro e o diestro, por meio de escova ginecológica e lavado uterino de baixo volume. Com esse ensaio, observou-se que não houve diferença significativa (P>0,05) no resultado da citologia uterina entre as diferentes técnicas avaliadas. No entanto, quando comparadas as fases do ciclo estral, houve uma diferença significativa (P<0,05) quanto ao percentual médio de polimorfonucleares neutrófilos, sendo de 8,07±2,80 no estro e de 1,55±0,28 no diestro. Conclui-se, desse modo, que as técnicas estudadas apresentaram resultados igualmente eficazes para detecção de neutrófilos ao exame citológico do útero em ambas as fases avaliadas do ciclo estral, apesar de, na fase de estro, a detecção de maior percentual de neutrófilos possibilitar um diagnóstico e um prognóstico mais precoces de endometrite e, portanto, uma conduta terapêutica mais adequada.(AU)


The endometrial cytology consists in an efficient and practical method for the diagnosis of uterine disorders in mares, contributing for therapeutic conduct and prognostic evaluation regarding the females' fertility. Therefore, the aim of this study was to compare different techniques of material sampling for endometrial cytology in mares, in order to verify possible qualitative and quantitative differences in the practicality of implementation of this methodology. Twelve multiparous mares underwent an endometrial cytological exam, during estrus and diestrus, by means of a cytobrush and a low-volume uterine flush. With this test, no statistical difference was observed (P>0.05) regarding the result of endometrial cytology between the evaluated techniques. However, in comparison to the estrous cycle's phases, a significant difference was verified (P<0.05) on the average percentage of polymorphonuclear neutrophils: 8.07±2.80 in estrus and 1.55±0.28 in diestrus. Thus, we conclude that the studied techniques presented equally effective results for the detection of neutrophils in the cytological exam for both evaluated stages of the estrous cycle, whereas in estrus phase, the detection of a higher percentage of neutrophils allows a more precocious diagnostic and prognostic of endometritis and, therefore, a more adequate therapeutic approach.(AU)

ABSTRACT

Opinions on ovariohysterectomy (OHE) of bitches vary depending on region and country. In this descriptive, prospective cross-sectional study, uterine tracts and ovaries exhibiting gross pathologic findings (n = 76) were collected post-surgery from a reference population of 3,600 bitches (2.11% incidence) that underwent elective OHE during September to November 2013 and evaluated by histopathology examination. Data were evaluated by using descriptive statistics and chi-squared tests. Bitches were of crossbred background with average age 5 years (range 0.6–8.0 years) and most were nulliparous (69.7%) with no anamnesis of reproductive diseases (81.6%). Frequencies of proestrus, estrus, and diestrus were 42.1%, 6.6%, and 19.7%, respectively. The presence of mammary gland masses (5.3%) significantly correlated with histopathologic findings in ovaries and age of the bitch (p < 0.05). Predominant uterine histopathologies included cystic endometrial hyperplasia, periglandular fibrosis, lymphoplasmocytary endometritis, and adenomyosis (19.7%, 14.5%, 4.0%, and 2.6%, respectively). In ovaries, hyperplasia of rete ovarii, follicular cysts, oophoritis, adenoma of the rete ovarii, cysts of superficial structures, and granulosa cell tumors (10.5%, 10.5%, 7.9%, 4.0%, 2.6%, and 2.6%, respectively) were observed. The results reveal the presence of subclinical pathologies in healthy bitches, suggesting that OHE at an early age is beneficial for prevention of reproductive pathologies.

ABSTRACT

O estudo das interações orgânicas da gestação e as mudanças fisiológicas que estão ocorrendo nesta fase são de extrema importância para a avaliação clínica da fêmea gestante ou para estabelecer o diagnóstico de processos patológicos em andamento. O objetivo do presente estudo foi comparar o perfil das diversas proteínas sanguíneas (frações protéicas do soro - albumina, a1, a2, b, g globulinas e proteína total) durante o período gestacional e no diestro em cadelas. Foram utilizadas 40 fêmeas caninas da raça Dogue Alemão, em idade variando entre 2 a 7 anos, clinicamente saudáveis. Os animais foram separados em dois grupos, denominados Grupo não gestante (NG) e Grupo gestante (G), constituídos por 20 fêmeas em diestro e 20 fêmeas gestantes, respectivamente. Preconizou-se colheita de sangue semanalmente de cada animal durante 9 semanas. Nas fêmeas do grupo NG, as amostras foram colhidas a partir do início do diestro, até a detecção do início do anestro; no grupo G, as amostras foram colhidas do início do diestro até o momento da parição. A partir do soro sanguíneo, foram determinadas as concentrações de proteína total, albumina, a1, a2, b e g globulinas. Não houve diferença significativa quanto aos resultados de proteína sérica total entre a 1ª e 6ª semanas de gestação e diestro, havendo decréscimo gradual em ambos grupos. Já ao final da gestação (entre a 7ª e 9ª semanas), houve acréscimo significativo dos valores de proteína total, sugerindo ação anabólica. Os níveis de albumina sofreram queda da 1ª a 9ª semana, tanto no grupo gestante, como não-gestante, com diferença estatística entre os dois grupos na 7ª, 8ª e 9ª semanas. As concentrações de α1 globulina nas fêmeas gestantes sofreram acréscimo significativo a partir da 2ª semana, contudo, diferença estatística entre os grupos NG e G ocorreu somente na 8ª e 9ª semanas, coincidindo com a fase de preparação à parição. Houve aumento no perfil de α2 globulina entre a 2ª e 3ª semanas de gestação, porém tais valores não diferiram das fêmeas em diestro. Diferença significativa de α2 globulina foi observada apenas durante a 4ª, 5ª, 6ª, 8ª e 9ª semanas entre os dois grupos avaliados. As concentrações séricas de ß globulina nas cadelas não-gestantes foi superior às das fêmeas gestantes durante a 2ª, 4ª e 7ª semanas. Durante o primeiro terço da gestação (1ª a 3ª semana), foi observado queda dos valores de γ globulina, coincidente ao período pré implantacional e durante a formação dos sítios de implantação. Entre a 8ª e 9ª semanas de gestação, houve acréscimo significativo de γ globulina, possivelmente conseqüente ao aumento da produção de imunoglobulinas direcionadas à glândula mamária, como constituinte do colostro. Em conclusão, as proteínas alteram-se de forma evidente durante o período de gestação. Foi possível inferir diferenças nas funções biológicas das proteínas sanguíneas em cadelas gestantes e não gestantes. As proteínas determinadas estão envolvidas com o estímulo inflamatório durante a gestação, além dos mecanismos de regulação hormonal e preparação do organismo materno à lactação.(AU)


The study of the organic interaction and physiological adaptations during pregnancy is of utmost importance for clinical evaluation and diagnosis of pathological conditions of pregnant bitches. The aim of the present study was to compare the serum protein profile (total protein, albumin, a1, a2, b and g globulin) of pregnant and diestrous bitches. For this purpose, 40 healthy 2 to7-year-old Great Dane bitches were used. The bitches were allocated in two experimental groups: Non-pregnant group (NP; n=20) and pregnant group (P; n=20). From each female, blood was drawn weekly during 9 weeks, from the diestrous onset until the beginning of anestrus or parturition, respectively from NP and P groups. The concentration of total protein, albumin and a1, a2, b and g globulins were determined from serum samples. No statistical difference was found for total protein between 1st and 6th weeks of pregnancy or diestrus. In both groups, there was a progressive decline in total protein concentration. At the end of pregnancy (7th to 9th week), a significant increase in total protein was verified, suggesting an anabolic process. Albumin concentration decreased between the 1st and 9th week in both groups, however, more markedly in the P group (with significant difference between groups at 7th, 8th and 9th week). The levels of α1 globulin in pregnant bitches increased significantly from the 2nd week on. Statistical difference was observed between groups only at the 8th and 9th week, during which a preparatory phase for parturition occurs. A significant rise in α2 globulin was shown between the 2nd and 3rd week of gestation, however without difference from the NP group. There was significant difference for α2 globulin between groups at the 4th, 5th, 6th, 8th and 9th week. Serum concentration of b globulin in diestrous bitches was superior than pregnant bitches at the 2nd, 4th and 7th week. During early gestation (1st to 3rd week) there was a decrease in g globulin, simultaneously to the preimplantation period and formation of implantation sites. An increase in g globulin was shown between the 8th and 9th week in pregnant bitches, possibly due to the increase in immunoglobulin synthesis targeting the mammary gland to form the colostrum. In conclusion, markedly changes in protein profile occur during gestation. It was possible to state different biological function of blood proteins in pregnant and diestrous bitches. The determined proteins are enrolled in the inflammatory stimulus during gestation, as well as in the hormonal regulatory mechanisms and maternal preparatory process to lactation.(AU)

ABSTRACT

The aim of this study was to evaluate the endometrial inflammatory response of nine Zebu cows inoculated through uterine infusion with 30 mL of inactivated Escherichia coli suspension (1 x 109 UFC/mL) and nine with 30 mL of sterile phosphate buffered saline. Endometrial biopsies were performed before and after the inoculations during estrus, and ten days later in the diestrus phase. Neutrophilic infiltrates were observed in 88.8% of samples from the E. coli group in estrus phase and demonstrated different degrees of endometrial inflammation. This study characterizes a suitable model for studying endometritis in cattle.(AU)

ABSTRACT

A primary characteristic of autism, which is a neurodevelopmental disorder, is impaired social interaction and communication. Furthermore, patients with autism frequently show abnormal social recognition. In mouse models of autism, social recognition is usually assessed by examining same-sex social behavior using various tests, such as the three-chamber test. However, no studies have examined the ability of male mice with autism to recognize the estrous cycle of female partners. In this study, we investigated the sexual behaviors, especially mounting and ultrasonic vocal communication (USV), of BTBR T+ tf/J (BTBR) mice, which are used as a well-known mouse model of autism, when they encountered estrus or diestrus female mice. As expected, C57BL/6 mice mounted more female mice in the estrus stage compared with the diestrus stage. We found that BTBR mice also mounted more female mice in the estrus stage than female mice in the diestrus stage. Although the USV emission of male mice was not different between estrus and diestrus female mice in both strains, the mounting result implies that BTBR mice distinguish sexual receptivity of females.

ABSTRACT

Immunohistochemical expression of proliferating cell nuclear antigen (PCNA) was evaluated in the endometrium of mares during estrus and at early diestrus. Three samples were collected by endometrial biopsy from 10 mares, on estrus/ second day, in the ovulation day and seven days after the ovulation day. PCNA expression was high in luminal epithelium and low in endometrial glands on samples taken on estrus/second day and on the ovulation day (p 0.05). For samples collected on the seventh day following ovulation, the averaged PCNA immunostaining was higher in glandular epithelium (p 0.05). The study revealed that luminal epithelial cells exhibit higher proliferation during estrus and glandular epithelial cells exhibited higher proliferation during diestrus...


A expressão do antígeno núcleo celular proliferante (ANCP) foi avaliada no endométrio de éguas durante o estro e início do diestro. Em cada uma de dez éguas foram efetuadas biópsias do endométrio em três momentos dos respectivos ciclos reprodutivos: segundo dia do estro, dia da ovulação e sete dias após a ovulação. Nas amostras colhidas no segundo dia do estro e no dia da ovulação, a expressão do ANCP foi elevada no epitélio luminal e baixa nas glândulas endometriais (p 0,05). Nas amostras colhidas no sétimo dia após a ovulação, a média de ANCP imunologicamente corado foi maior no epitélio glandular (p 0,05). O estudo revelou que as células do epitélio luminal apresentaram a maior proliferação durante o estro e que as células epiteliais glandulares apresentaram a maior proliferação durante o diestro...

The pyometra is a disease that affects middle age and elderly female dogs during diestrus. Hormonal, microbiological, biochemical and hematological aspects are well described. However, few studies have evaluated the role of each in the prognosis of canine pyometra. The aim of this study was to identify markers associated with clinical worsening of dogs with pyometra. We prospectively evaluated 80 dogs with pyometra treated surgically. Group 1 consisted of dogs that were discharged within 48 hours after surgery and Group 2 consisted of those who required prolonged hospitalization or died. The findings of hematological, biochemical and blood lactate levels were compared between groups and variables such as bacterial multidrug resistance, systemic inflammatory response syndrome (SIRS), hyperlactatemia and increased creatinine were analyzed through the dispersion of frequencies between groups. Among the variables studied, the presence of SIRS and elevated serum creatinine >2.5mg/mL were effective in predicting the worsening of the disease and can be used as prognostic markers of canine pyometra...


A piometra é doença que acomete as fêmeas da espécie canina de meia-idade a idosa durante o diestro. Aspectos hormonais, microbiológicos, hematológicos e bioquímicos são bem descritos. Entretanto, são poucos os estudos que avaliam o papel de cada um desses no prognóstico da piometra canina. O objetivo deste estudo foi identificar marcadores associados ao agravamento clínico de cadelas com piometra. Foram avaliadas prospectivamente 80 cadelas com piometra tratadas cirurgicamente. O grupo 1 foi composto por cadelas que receberam alta até 48 horas de pós-operatório, e o grupo 2 por aquelas que necessitaram de internamento prolongado ou morreram. Os achados hematológicos, bioquímicos e os níveis do lactato sanguíneo foram comparados entre os grupos, e variáveis como multirresistência bacteriana, síndrome da resposta inflamatória sistêmica (SIRS), hiperlactatemia e aumento da creatinina foram analisadas por meio da dispersão de frequências entre os grupos. Entre as variáveis estudadas, a presença de SIRS e a elevação da creatinina sérica >2,5mg/mL foram eficazes em predizer o agravamento da doença e podem ser utilizadas como marcadores prognósticos da piometra canina...

ABSTRACT

Este trabalho leve o objetivo de avaliar a influência do EGF na maturação in vitro (MIV) de cadelas. Realizou-se o transporte dos ovários em solução de cloreto de sódio 0,9%, que foram seccionados em solução salina fosfato temponado (PBS) e os complexos cumulus oócitos (COCs) selecionados em meio de cultura de tecidos (TCM) 199 suplementado com HEPES. Foram coletados 405 oócitos grau 1 de cadelas em anestro e diestro. Os oocitos provenientes das duas fases do ciclo estral foram submetidos a dois tratamentos: meio com adição de 10ng/mL do fator de crescimento epidermal (EGF) (T) e meio sem suplementação (C). Depois de 72 horas de maturação, os COCs foram desnudados, fixados e corados com HOESCHT 33342 para avaliação da maturação nuclear. Os oócitos obtidos dos ovários da fase de diestro do grupo T demonstraram maior porcentagem (18,8%) de oócitos no estágio de metáfase II (M-II), em relação ao grupo C (1,3%) (p < 0,01). Nos oócitos obtidos da fase de anestro houve diferença (p < 0,05) entre os grupos, observando-se menor parcela (4,1%) de oócitos no estágio de vesícula germinativa (VG) no grupo T, quando comparado ao grupo C (16,1%). Comparando-se as diferentes fases reprodutivas, em meios suplementados com EGF, foi observada diferença (p < 0,05) apenas nos oócitos obtidos da fase de diestro em relação ao estágio de M-II. Dessa maneira, a suplementação no meio de maturação na concentração de 10 ηg/mL, neste estudo, exerceu influência positiva apenas na MIV de oócitos caninos oriundos da fase de diestro, incrementando os índices de M-II nessa espécie.


This work aimed to evaluate the influence of EGF on canine oocyte in vitro maturation (IVM). We carried out the transport of the ovaries in sodium chloride 0.9% solution, which were cut into phosphate-buffered saline (PBS) and the cumulus oocyte complexes (COCs) selected in tissue culture medium (TCM), supplemented with 199 HEPES. A total of 405 grade 1 oocytes were collected from bitches in anestrus and diestrus. Oocytes from the two phases of estrous cycle were subjected to two treatments: medium with addition of 10 ηg/mL epidermal growth factor (EGF) (T) and medium without supplementation (C). After 72 h of maturation, COCs were denuded, fixed and stained with Hoechst 33342 to assess nuclear maturation. The oocytes obtained from the ovaries of the diestrus phase of the T group demonstrated higher percentage (18.8%) of oocytes at metaphase II stage (M-II) than C group (1.3%) (p < 0.01). The oocytes from anestrus phase demonstrated difference (p < 0.05) between the groups, observing smaller proportion (4.1%) of oocytes at the stage of germinal vesicle (GV) in group T compared to group C (16.1%). Comparing the different reproductive status in media supplemented with EGF difference (p < 0.05) was observed only in oocytes diestrus phase relative to the stage of M-II. Thus, supplementation on maturation medium at a concentration of 10 ηg/mL of EGF in this study had positive influence only on IVM of canine oocytes obtained from diestrus phase, increasing the levels of M-II in this species.

ABSTRACT

In canine specie, oocyte maturation rates are low and the percentage of oocytes that remain in the stage of germinal vesicle (GV) regardless of culture conditions is high. During maturation oocyte undergoes modification and the GV chromatin remodeling manifested by changes in the configuration and positioning. The objective of this work is to evaluate the configuration and positioning of chromatin of oocytes in GV stage during anestrus and diestrus bitches. The ovaries of 33 females (20 bitches in anestrous and 13 in diestrus) were isolated, sliced and only cumulus-oocyte complexes (COCs) grade 1 were subjected to solution of 0.2% hyaluronidase for release in cumulus cells. After this process, the selected oocytes were stained and evaluated. From a total of 920 oocytes, 566 were classified as grade 1 and the stages of chromatin configuration identified as GV-1, GV-2, GV-3 and GV-4. The observed changes in chromatin configuration been characterized as a transition dispersed chromatin (GV-1, GV-2) for partially condensed (GV-3) until it reaches a fully condensed stage (GV-4). The data analyzed from the chromatin configuration showed a significant difference between the stages with a higher proportion of GV-1 and GV-2 for the anoestrus and GV-3 and GV-4 during diestrus. Na espécie canina as taxas de maturação oocitária são baixas e a porcentagem de oócitos que permanecem em estagio de vesícula germinativa (VG), independente das condições de cultivo, e alta. Durante a maturação oocitária, a VG sofre modificação e remodelamento da cromatina, que se manifesta por alterações na sua configuração e posicionamento. Assim, o objetivo deste trabalho e avaliar a configuração e o posicionamento da cromatina de oócitos em estagio de VG durante o anestro e diestro de cadelas. Os ovários de 33 fêmeas (20 cadelas em anestro e 13 em diestro) foram isolados, fatiados e os complexos cumulus-oócitos (COCs) foram submetidos à solução de hialuronidase 0,2% para a liberação das células do cumulus. Apos esse processo, os oócitos selecionados foram corados, avaliados e apenas COCs grau 1 foram utilizados. De um total de 920 oócitos, 566 foram classificados como grau 1 e os estágios de configuração da cromatina identificados como VG-1, VG-2, VG-3 e VG-4. As alterações observadas na configuração da cromatina foram caracterizadas como transição de uma cromatina dispersa (VG-1, VG-2) para parcialmente condensada (VG-3) ate atingir um estagio totalmente condensado (VG-4). Os dados analisados da configuração da cromatina mostraram uma diferença significativa entre as fases de anestro e diestro, com maior proporção de VG-1 e VG-2 durante o anestro e de VG-3 e VG-4 durante o diestro. Ha necessidade de novos estudos para uma compreensão adequada da influencia da configuração da cromatina de oócitos no estagio de VG na retomada da meiose e na competência meiótica do oócito...

ABSTRACT

No ciclo estral de cadelas a fase luteínica, denominada diestro, compreende um período que varia de 60 a 100 dias em animais não-prenhes, caracterizado pela elevação plasmática de progesterona nos primeiros 20 dias pós ovulação (p.o). A adiponectina é a mais abundante proteína secretada pelo tecido adiposo, porém sua concentração plasmática diminui significativamente em alterações metabólicas como resistência insulínica e Diabetes mellitus tipo2, alterações descritas como relacionadas em algumas cadelas com o período de diestro. O objetivo do estudo foi determinar a expressão e imunolocalização do sistema adiponectina (adiponectina e seus receptores, adipoR1 e adipoR2) no corpo lúteo de cadelas ao longo do diestro, correlacionando-o ao perfil hormonal de 17β-estradiol e progesterona, assim como à expressão de um dos genes alvo do sistema, o PPAR-γ. Para realização do estudo foram coletados corpos lúteos de 28 cadelas durante ovariosalpingohisterectomia de eleição nos dias 10, 20, 30, 40, 50, 60 e 70 pós ovulação (o dia zero da ovulação foi considerado aquele no qual a concentração plasmática de progesterona atingiu 5ng/mL). Os corpos lúteos foram avaliados por imunohistoquímica para adiponectina e seus receptores e a expressão do RNAm do PPAR-γ por PCR em tempo real. A análise estatística da avaliação gênica foi realizada com o teste ANOVA, seguido por comparação múltipla Newman-Keuls. O sinal da adiponectina apresentou-se mais intenso até os primeiros 20 dias p.o, momento de regência da progesterona; houve queda gradativa após este período, coincidindo com a ascensão do 17β-estradiol, cujo pico foi notado próximo do dia 40 p.o. A queda marcante da adiponectina ocorreu após 50 dias p.o. O sinal do adipoR1 mostrou-se bem evidente até os 40 dias p.o e o do adipoR2 até os 50 dias p. o, decaindo posteriormente. Foi observada maior expressão do gene PPAR-γ aos 10, 30 e 70 dias p.o. Estes resultados mostram que a expressão protéica da adiponectina e de seus receptores se altera ao longo do diestro e que estas alterações podem estar relacionados às alterações hormonais e expressão do PPAR- γ, participando do mecanismo fisiológico de desenvolvimento, manutenção, atividade e regressão luteínica em cadelas.


In the estrous cycle of bitches, the luteal phase or diestrus includes a period ranging from 60 to 100 days in non-pregnant animals, characterized by elevated serum progesterone during the first 20 days post-ovulation (p.o). Adiponectin is the most abundant protein secreted by adipose tissue, but plasma concentration decreases significantly in metabolic disorders like insulin resistance and diabetes mellitus type 2, described as related changes in some bitches in diestrus. The aim of this study was to determine the expression and immunolocalization of the adiponectin system (adiponectin, and adipoR1 adipoR2) in the corpus luteum during diestrus, and correlate it to hormonal profile of 17β-estradiol and progesterone, as well as the expression of a gene target of the system, the PPAR-γ. For the study, corpora lutea were collected from 28 dogs during ovariosalpingohysterectomy on days 10, 20, 30, 40, 50, 60 and 70 post ovulation (day zero of ovulation was considered the day when the plasma progesterone concentration reached 5ng/mL). The corpora lutea were evaluated by immunohistochemistry for adiponectin, adipoR1 and adipoR2 and mRNA expression of PPAR-γ by real-time PCR. Statistical analysis of gene expression was performed with ANOVA followed by Newman-Keuls multiple comparisons. Adiponectin positive signal was stronger during the first 20 days p.o, time of the regency of progesterone; there was a gradual adiponectin and progesterone decline after this period, coinciding with the rise of 17β-estradiol, whose peak was near the 40 days p.o. The markedly adiponectin decrease occurred after 50 days p.o. The signal of adipoR1 was markedly evident at 40 days p.o and that of adipoR2 up to 50 days p.o, declining afterwards. We observed higher expression of PPAR-γ gene at 10, 30 and 70 days p.o. These results show that adiponectin and its receptors protein expression is altered during the diestrus and that these changes may be related to hormonal changes and expression of PPAR-γ, participating in the physiological mechanism of development, maintenance, activity and luteal regression in bitches.

ABSTRACT

El cuerpo humano presenta partes anatómicas pares y simétricas de forma global. A pesar de la simetría, en su funcionamiento se utiliza preferentemente un lado frente al otro, concepto que corresponde al de lateralidad. La lateralización es la supremacía de un hemisferio cerebral sobre el otro, la preferencia del uso de un hemicuerpo sobre el otro. La dominancia es el predominio de un miembro del cuerpo sobre su simétrico, por ejemplo una mano sobre la otra. A veces, se observan patologías orales como caries y gingivitis en la cavidad bucal de un paciente, con predominancia en una hemiarcada. En este estudio se evaluó la distribución lateral de las patología orales y la predominancia de uso de una de las manos en 71 niños de 6 a 17 años que acudieron en la cátedra de odontopediatria e higiene de la Facultad de Odontología de la Universidad Nacional de Asunción como pacientes o acompañantes, en el mes de junio en el año 2012. Fue un estudio observacional descriptivo de corte transversal, con el componente analítico. Se observó en los pacientes si éstos eran diestros o zurdos, la presencia de caries, manchas blancas, dientes perdidos y obturados, y la técnica de cepillado. Al promediar el número de patologías por lado en la totalidad de los pacientes se encontró en el lado derecho 50,2% y en el lado izquierdo 49,8% no siendo significativas las diferencias según el lado. (P=0,954).

Objetivo: determinar as fases do ciclo estral em ratas, utilizando o lavado vaginal, fornecendo a pesquisadores considerações úteis para o seu reconhecimento. Métodos: coleta da secreção vaginal de ratas wistar, por lavado vaginal a fresco, observada em microscópio ótico com objetivas de 10 e 40 vezes, realizada pela manhã, durante dois ciclos completos, em 70 ratas. Resultados: o ciclo estral tem duração média de quatro dias e pode ser dividido em diestro I e II, proestro e estro, os quais podem ser determinados pela distribuição dos tipos celulares no lavado vaginal. Os três tipos celulares que predominam são os leucócitos, células arredondadas nucleadas e células epiteliais cornificadas. Conclusão: o lavado vaginal de ratas constitui um método importante para o estudo das alterações que ocorrem durante o ciclo estral, devido às mudanças abruptas nos níveis de estradiol que ocorrem nas quatro fases do ciclo. A observação é rápida e fácil de ser realizada.

ABSTRACT

PURPOSE: We established a rat model to investigate the female vaginal arousal response and the effects of the estrous cycle on vaginal blood flow in vivo. MATERIALS AND METHODS: Laser Doppler flowmetry was used to measure changes in vaginal blood flow induced by pelvic nerve stimulation(PNS) in 30 female Sprague-Dawley rats. Frequency response data were obtained in each animal. In addition, the rat's stage in the estrous cycle was determined according to the cell types observed in a vaginal smear. Changes in blood flow at 10 Hz frequency caused maximal response, and these responses were evaluated by comparing the relative peak flow, time to peak, duration of response, and relative area under the curve(AUC) according to the estrous cycle. RESULTS: Reproducible frequency dependent increases in vaginal blood flow were observed in response to PNS. Relative peak flow, time to peak, duration of response and relative AUC were slightly greater in proestrus and estrus groups(relatively higher estradiol level, n=17) than those in metestrus and diestrus groups(relatively lower estradiol level, n=13). However, these differences were not statistically significant. CONCLUSIONS: Our data suggest that the rat is a useful and reliable animal model for investigating the vaginal arousal response. In addition, the estrous cycle of the animal does not seem to be an important confounding factor in this animal model.

ABSTRACT

As a step towards the identification of genes preferentially expressed in the oviduct during early rat embryo development, we isolated a cDNA fragment (Pr14) by using RNA arbitrarily primed PCR (RAP-PCR), being its expression restricted to oviduct and uterus; its mRNA is mainly expressed in oviduct during late luteal phase and early pregnancy. This fragment is 100 per cent identical to a rat DNA sequence (Accession No. NW_047400)downstream the terminal exon of a Ratturs norvegicus gene (Locus Link Accession No. LOC289316) similar to ebaf (endometrial bleeding-associated factor), a novel member of the Transforming Growth Factor superfamily. Northern analyses showed that this sequence hybridizes with 2.9 kb and 4.1 kb mRNAs in early pregnant rat oviducts. However, only the 4.1 kb mRNA was detected in the oviduct of non-pregnant rats, showing an increase from proestrus to diestrus. The expression of this oviduct-uterus specific mRNA suggests that the products of this gene may play a role in the oviductal reproductive process.

ABSTRACT

BACKGROUND: Pituitary adenylate cyclase-activating polypeptide (PACAP) plays the role of a hypophysiotropic factor, which regulates the synthesis and secretion of pituitary hormones through the hypothalamo-hypophysial portal system. No clear evidence has yet been reported regarding the regulation of prolactin (PRL) by PACAP. In the present study, we tested a hypothesis that PACAP regulates the synthetic machinery of PRL during the estrus cycle and pubertal process using intracerebroventricular (i.c.v.) injection of an antisense oligodeoxynucleotide (ODN) against type I PACAP receptor (PAC1). METHODS: An RNase protection assay (RPA) was used to determine the pattern of hypothalamic PACAP and PAC1 mRNA expressions during the estrus cycle. Antisense PAC1 ODN was administered via i.c.v. injection to the female rats in normal estrus cycle of pubertal process. Northern blot analysis was used to determine the mRNA ievel of PRL in the pituitary gland. RESULTS: 1) PACAP mRNA in the medial basal hypothalamus was significantly increased at the diestrus I, while PAC1 mRNA showed no significant change. 2) PRL mRNA level of pituitary was increased by an injection of antisense PAC1 ODN at the proestrus and estrus stages. 3) PRL mRNA level of pituitary was significantly decreased by antisense PAC1 ODN injection at stage of prepuberty and initiate puberty, while its level was increased at stage of puberty. CONCLUSION: These data suggest that PACAP suppresses PRL mRNA synthesis through the PAC1 signaling pathway in the certain estrus cycle environments. It may be also involved in the regulation of pituitary PRL gene expression during the pubertal process

ABSTRACT

OBJECTIVE: This study was to investigate the expression of CDK inhibitors, p27(kip1) and p57(kip2) in mouse endometrium during the estrus cycle and pregnant period. METHODS: Total RNA and protein were extracted from endometrium of mouse sacrificed at diestrus, proestrus, estrus, and metestrus cycle, and at day 1-6 post-coitum (p.c.), then semi-quantitative RT-PCR and western blotting of p27(kip1) and p57(kip2) was carried out. RESULTS: p27(kip1) and p57(kip2) mRNA was highly expressed in diestrus and proestrus stage than estrus and metestrus stage. In comparison with estrus cycle, p27(kip1) and p57(kip2) mRNA level was highly maintained in gestational endometrium (except p27(kip1) of day 5 p.c). p57(kip2) protein level was relatively low from day 1 p.c. to day 4 p.c. But it was significantly increased in day 5 p.c. and day 6 p.c. CONCLUSION: These results show that p27(kip1) and p57(kip2) may play a role in endometrial differentiation for regular estrus cycle and implantation, and especially p57(kip2) may play an essential role in endometrial differentiation for maintenance of implantation.

OBJECTIVE: This study was to investigate the localization of CDK inhibitor, p57(kip2) in mouse endometrium during the estrus cycle and pre- and peri-implantation periods. METHODS: The p57(kip2) protein was immunostained from endometrium of mouse sacrificed at diestrus, proestrus, estrus, and metestrus cycle, and at day 1-6 post-coitum (p.c.). RESULTS: The staining in the luminal epithelium was very weak in comparison with glandular and stromal cells. In diestrus stage, immunoreactivity of p57(kip2) was heterogeneously strong in parts of decidualized or degenerated stromal cells. In proestrus stage, strong immunoreactivity p57(kip2) was largely found in stromal cells. But, p57(kip2) was showed low immunoreactivity in estrus stage. In metestrus stage, immunoreactivity of p57(kip2) was heterogeneously strong in decidualized stromal cells. In day 1-2 p.c., immunoreactivity of p57(kip2) was low in some endometrial stromal cells. In day 3-4 p.c., immunoreactivity of p57(kip2) was strong in some endometrial stromal cells. In day 5-6 p.c., immunoreactivity of p57(kip2) was strong in decidual cells. CONCLUSION: These suggest that p57(kip2) may play an essential role in endometrial differentiation for maintenance of implantation, especially decidualization of endometrial stromal cells.

ABSTRACT

OBJECTIVE: Bok, Bcl-2-related ovarian killer, is a proapoptotic Bcl-2 family protein identified in the ovary based on its dimerization with the antiapoptotic protein Mcl-1. The present study examined the hormonal regulation and localization of Bok messenger RNA levels in the rat ovary during the follicle development. METHODS: We used the female rats of SD strain. Bok mRNA levels in the ovary were determined by Northern blot analysis. In situ hybridization were performed to determine the specific ovarian cell type expressing Bok mRNA. RESULTS: Northern blot analysis of ovaries obtained from immature rats revealed increasing levels of Bok mRNA during postnatal development. The major cell types expressing Bok mRNA were the granulosa cells of preantral and atretic follicles. Treatment of immature rats with diethylstilbestrol (DES) for 24-48 h increased ovarian Bok mRNA levels. Bok mRNA was remained the same levels in rats removed DES for 24- 48 h to induce apoptosis. High signals of Bok mRNA after DES treatment were detected in granulosa cells of early antral follicles. Treatment of immature rats with pregnant mare's serum gonadotropin (PMSG) for 12 h increased markedly ovarian Bok mRNA expression which was detected mainly in preantral and atretic follicles. Interestingly, low levels of Bok mRNA were also expressed in granulosa cells of preovulatory follicles. Treatment of PMSG-primed rats with human chorionic gonadotropin (hCG) stimulated strongly ovarian Bok mRNA expression at 6-9 h. At that time, Bok mRNA was expressed in granulosa cells of atretic and small growing follicles. In adult estrus cyclic ovaries, Bok gene expression was higher on proestrus and estrus than metaestrus and diestrus. Moreover, the highly increased expression of Bok mRNA was found in rat ovaries at 48 h after hypophysectomy. CONCLUSION: These results demonstrate that Bok is one of proapoptotic Bcl-2 members expressed in early growing and atretic follicles during the ovarian follicular development. Gonadotropins induce a transient increase of Bok gene expression in granulosa cells of preantral and preovulatory follicles indicating some role in the ovulatory process.
